چکیده

Foci of p53 binding protein 1 (53 BP1) have been used as a biomarker of DNA double-strand breaks (DSBs) in cells induced by ionizing radiations. 53 BP1 was shown to relocalize into foci shortly after irradiation, with the number of foci closely paralleling the number of DNA DSBs. However, consensus on criteria in terms of the numbers of 53 BP1 foci to define cells damaged by direct irradiation or by bystander signals has not been reached, which is partly due to the presence of 53 BP1 also in normal cells. The objective of the present work was to study the changes in the distribution of cells with different numbers of 53 BP1 foci in a cell population after low-dose ionizing irradiation (o0.1 Gy) provided by alpha particles, with a view to propose feasible criteria for defining cells damaged by direct irradiation or by bystander signals. It was proposed that the change in the percentage of cells with 1–3 foci should be used for such purposes. The underlying reasons were discussed. & 2011 Elsevier B.V. All rights reserved.

P53 Binding Protein 1 (53bp1) Is an Early Participant in the Cellular Response to DNA Double-Strand Breaks

p53 binding protein 1 (53BP1), a protein proposed to function as a transcriptional coactivator of the p53 tumor suppressor, has BRCT domains with high homology to the Saccharomyces cerevisiae Rad9p DNA damage checkpoint protein. To examine whether 53BP1 has a role in the cellular response to DNA damage, we probed its intracellular localization by immunofluorescence. In untreated primary cells a...

Evaluating Gamma-H2AX Expression as a Biomarker of DNA Damage after X-ray in Angiography Patients

Objective: Coronary heart disease (CHD) is one of the most common diseases. Coronary angiography (CAG) is an important apparatus used to diagnose and treat this disease. Since angiography is performed through exposure to ionizing radiation, it can cause harmful effects induced by double-stranded breaks in DNA which is potentially life-threatening damage. The aim of the present study is to inves...

Modeling the distribution of deposited energy by alpha particles from Radon 223 decay and its effect on DNA

The ionizing radiations, through physical and chemical processes, lead to simple and complex single- and double- strand breaks, as well as base lesions to the DNA. In this study, taking into account all the physical and chemical processes involved in the interaction of ionizing radiation with matter, the initial damage induced to DNA was evaluated for 5.7 MeV alpha-rays from Radon 223 isotope....

DNA double strand breaks do not play a role in heat-induced cell killing.

In their recent article, Takahashi et al. (1) suggest that heatinduced DNA double strand breaks contribute to heat-induced cell killing because heat treatment induces histone gH2AX-containing foci. Such foci have been associated with double strand breaks induced by ionizing radiation, other agents, and other stresses (2). However, the authors disregard the hyperthermia biology literature, which...
